Can DNA be extracted from dried blood?

It is possible to extract DNA from dried blood. The dried blood is collected using a sterile swab, which has been moistened with sterile water. The swab is submitted for DNA extraction.

Is it possible to determine if two blood samples collected from different areas belong to the same individual?

Every individual has a unique DNA profile. Thus using DNA profiling it is possible to determine if the blood samples belong to the same or to different individuals. If the DNA profile of one blood sample matches exactly to the DNA profile obtained from the other blood sample, this means that the blood sample comes from the same individual. ONLY IDENTICAL TWINS have the same DNA profile.

Is it possible to distinguish between blood and a red paint?

It is possible to determine if a red/ reddish brown stain is blood or not. A presumptive test is carried out. This test gives a characteristic greenish blue colouration if blood is present but not if the stain is paint or rust etc.

Is it possible to determine the presence of semen on clothes/car seats etc.?

It is possible. Semen stains on clothes etc. are easily visible under UV light. Following the detection of the stain, a presumptive test for the presence of semen I carried out. This test gives a purplish pink colouration if acid phosphatase (enzyme present in semen) is present in the stain.

From what samples is it possible to extract DNA?

Human DNA can be extracted from any body fluid or tissue (saliva, semen, blood, hair, nails etc.) Also DNA can be extracted from any item that is contaminated with body fluids (chewing gum, cigarette butts, sanitary towels etc.)

Is it possible to determine if 2 males having the same surname belong to the same paternal lineage?

This is possible since, the Y chromosome (part of DNA that determines male sex) is inherited from father to son. The regions of Y chromosome tested remain the same for over 10 generations. Thus if 2 males having the same surname and have the same Y chromosome means that they have the same ancestor.
